Yes, this is possible...with the notable exception of time tracking and expense reports unfortunately. Those are hardcoded into the system, so to modify them you would have to use the Excel Addin. In the future all of the reports will be editable.
Creating a new report from a System report?
Is it possible to use a System report as the basis for a new report? In other words, I want to take a Time Tracking report, but I don't want some of the columns. I want to create a new report without starting from scratch.

Josh Santos, Two years ago you mentioned "In the future all reports will be editable" and was wondering if using system reports as base for other reports is even in your enhancement queue?
I would like to use 'Resource Load per Task' as the basis for a new report with an additional column for email address. Above, it is mentioned that only Time Tracking and Expense reports are not editable but I can't figure out how to do this with Resource Load report either. Any I missing something?
